These pathways encompass nociceptive TRPV1 hyperpolarization, CB1 and CB2 receptor activity, and the interaction with mu-opioid receptors.

Leveraging the power of Canalytics’ proprietary algorithms to process Certificate of Analysis (COA) data, we calculate the suggested synergistic effects and strengths of cannabinoids and terpenes involved in pain relief.

Effects of Terpenes on Nociception TRPV1 Signal Hyperpolarization for Pain Relief

Mu Opioid Receptor Activation: Beta-Caryophyllene’s pain-relieving effects are synergistically contributed by its activity on MU Opioid and CB2 receptors. The validation of its opioid effect pathway is reinforced by the effective blockade of a significant portion of Beta-Caryophyllene’s pain relief by opioid receptor antagonists (blockers) like Naloxone.

Direct Nociceptive Action: CB1 receptor activation triggers the opening of potassium (K+) channels in the nociceptor cell membrane. This mechanism raises the threshold necessary for pain signal generation through nociceptive hyperpolarization, effectively diminishing the probability of pain signals being generated by the nociceptive nerves. As a result, the likelihood of experiencing pain sensations is significantly reduced.

Nociceptive TRPV1 Hyperpolarization: The primary terpene, Myrcene, along with secondary terpenes, Nerolidol and Humulene, exhibit the greatest effects on nociceptive TRPV1 hyperpolarization. Our cannabis analysis algorithm for comfort and pain assesses cannabis COA samples based on the expected synergistic activity of CB1, CB2, TRPV1, and Mu Opioid receptors.

Peripheral Nociceptive Action: CB2 Ca+ signaling actively reduces the release of activators and sensitizers, thereby contributing to pain modulation. The endocannabinoid system, located at the peripheral terminal of the nociceptive receptor, serves as the body’s initial endogenous defense against pain and can be activated through CB1 or CB2 receptor stimulation.
